Dr. Liang Lingmin , senior researcher at Jinfeng Laboratory and technical director of the Stem Cell Center Resource Library. He has long been engaged in clinical-grade stem cell resource construction, differentiation regulation and cell therapy research. Established multiple clinical-grade stem cell lines and achieved functional differentiation, supporting the first domestic clinical research on human embryonic stem cells to treat Parkinson's disease and obtaining multiple IND approvals. Participate in the formulation of the first international, national and group standards for stem cells to help standardize the development of the industry 。

Dr. Liang Lingmin introduced the construction of the National Stem Cell Resource Bank (Chongqing) branch. This branch is the first branch of the National Stem Cell Resource Bank approved by the Ministry of Science and Technology and the Ministry of Finance. It undertakes the three major tasks of backup of the national general library in the west, regional benchmarking and resource sharing in the west. It focuses on the three major directions of physiological, pathological and special environment stem cells. It has a storage capacity of 2 million copies and an ISO 20387+ISO 17025 dual-accredited quality system. With the launch of the first stem cell drug, the official implementation of the "Regulations on the Administration of Clinical Research and Clinical Translation Application of New Biomedical Technologies", and the FDA's listing of organoids as an alternative to animal experiments, standardized resource libraries have become a key infrastructure connecting basic research and clinical translation. Dr. Yao Junjun , a young scholar from Jinfeng Laboratory. Mainly engaged in research on stem cell biology and regenerative medicine, he has accumulated systematic and in-depth technical experience in the isolation and culture, functional analysis and transformation application of neural stem cells, mesenchymal stem cells and microglia. For the first time, the molecular profile of human hippocampal neural stem cells in aging and damaged states was systematically analyzed and a new type of microglia was discovered. 。
Key points of the lecture
Dr. Yao Junjun took the team's newly discovered microglia subpopulation OLM as an example to demonstrate the research path from basic research to clinical translation. Most neurodegenerative diseases are accompanied by demyelinating damage, which cannot be reversed by existing treatments. The fundamental reason is that the myelin repair mechanism is unknown. The team integrated single-cell nuclear transcriptome data from multiple brain regions in patients with multiple neurodegenerative diseases and identified oligodendrocyte-like microglia (OLM). While retaining the identity of microglia, this subtype endogenously expresses key genes of the oligodendrocyte lineage, is conserved across species, and has close spatial proximity and ligand-receptor interactions with oligodendrocytes. Functionally, OLM can significantly promote myelin repair in progeria SAMP8 mice and spinal cord injury rats, providing a new endogenous cell resource for cell therapy of demyelinating diseases. 。

For biological drugs, immunogenicity is a systemic risk caused by the interaction of drugs, populations, individual immune status and disease background, which directly affects drug exposure, efficacy and safety.
